天才少年理查德（邁爾斯·特勒 Miles Teller 飾）憑藉物體傳送裝置的發明得到正研究量子門的富蘭克林•斯通的賞識，他將理查德招至麾下，通過和維克多（託比·凱貝爾 Toby Kebbell 飾）、約翰尼（邁克爾·B·喬丹 Michael B. 　　四年前的紐約市，蟑螂擴散的病毒造成瘟疫，差點將市內兒童滅絕，在公立學校任教的昆蟲專家李美（艾莉絲高朗士Alix Koromzay飾）想出以毒攻毒方法：利用螞蟻及白蟻的DNA研製一種取名「猶太」的新品種蟑螂，藉著「猶太」體內的特殊分泌消滅蟑螂的體液，利用它們充當誘餌出賣肆虐的同類，「猶太」潛入蟑螂邋遢的棲息地把帶菌蟑螂一網打垮，病毒最後亦得以控制。　　破壞計劃看似空前成功，人類在四年後的今天還以為頑固的蟑螂絕跡人間，可是，並非所有「猶太」的也在預期中百多日壽命限期內死亡，部份變種成新種類，這種超智版蟑螂能模仿人類，牠們殺害人類後，把受害者的麵皮戴上，穿上大褸及利用兩條後腿走路，牠們在李美的殘破學校及黑暗的地鐵管道匿藏。　　與此同時，多宗殘酷兇殺案的死者均是李美的的前男友，警方認為李美的嫌疑最大，遂派遣自以為聰穎過人的探員卡拉斯基\對李美進行調查。卡拉斯基起初也對李美充滿懷疑，但他卻不相信身型普通的李美，有足夠力氣在殺人後撕走前男友的麵皮，再把屍體掛上高架燈柱之上。史上最巨大的無敵蟑螂再度變成殺人怪物，肯定對全人類安全構成威脅，李美只好再次迎戰！
